چکیده

Inbred male Lewis (LEW) and Brown Norway (BN) rats were used for experiments. Plastic dish nonadherent bone marrow cells (BMC) were prepared and cultured with 2% PWM-SCM for 6 days in a fibrin clot culture system to detect colony-forming units in culture (CFU-C). Clusters (10 to 50 cells per aggregate) and colonies (>50 cells per aggregate) in cultures were counted as CFU-C. LEW-derived CFU-C was identified by in situ immuno· histochemical staining of cultured cells in the fibrin clot, using a standard three-step avidin-biotin complex method with monoclonal antibody L21-6. which recognizes the class II MHC antigens of LEW but not BN rats.

Donor hematopoietic progenitor cells in nonmyeloablated rat recipients of allogeneic bone marrow and liver grafts.

BACKGROUND Although the persistence of multilineage microchimerism in recipients of long-surviving organ transplants implies engraftment of migratory pluripotent donor stem cells, the ultimate localization in the recipient of these cells has not been determined in any species. METHODS Progenitor cells were demonstrated in the bone marrow and nonparenchymal liver cells of naive rats and in Bro...

Expansion of Non-Enriched Cord Blood Stem/Progenitor Cells CD34+ CD38- Using Liver Cells

Many investigators have used xenogeneic, especially murine stromal cells and fetal calf serum to maintain and expand human stem cells. The proliferation and expansion of human hematopoietic stem cells in ex vivo culture were examined with the goal of generating a suitable protocol for expanding hematopoietic stem cells for patient transplantation. Using primary fetal liver cells, we established...

GENE THERAPY Transduction of donor hematopoietic stem-progenitor cells with Fas ligand enhanced short-term engraftment in a murine model of allogeneic bone marrow transplantation

Fas-mediated apoptosis is a major physiologic mechanism by which activated T cells are eliminated after antigen-stimulated clonal expansion generates a specific cellular immune response. Because activated T cells are the major effectors of allograft rejection, we hypothesized that genetically modifying allogeneic bone marrow (BM) cells prior to transplantation could provide some protection from...

Liver chimerism after allogeneic blood stem cell transplantation.

UNLABELLED Blood stem cells can mature into elements of many different lineages. We investigated the presence and nature of donor-derived (chimeric) cells within the liver after allogeneic stem cell transplantation. METHODS Liver biopsy autopsy specimens were examined from nine female patients who had undergone allogeneic bone marrow (n = 6) or peripheral stem cell (n = 3) transplantation fro...

Advances in Hematopoietic Stem Cell Mobilization and Peripheral Blood Stem Cell Transplantation

Hematopoietic stem/progenitor cells (HSPCs) which give rise to different blood cell types are present within the bone marrow microenvironment, especially in flat bones such as skull, vertebrae, pelvis and chest. Interacting factors such as stromal derived factor-1/CXCR4, very late antigen-4/vascular cell adhesion molecule-1, Lymphocyte function-associated antigen-1/ intercellular adhesion molec...
